Antibody-drug conjugates (ADCs) combine the targeting capabilities of monoclonal antibodies with the potent cytotoxic effects of small molecule drugs, offering precise targeting of cancer cells while minimizing off-target effects.

Modular Linker Design for Precision Payload Delivery

SpiroChem has established a strong expertise in linker-payload design, including a broad range of bioconjugation anchors and both chemically and enzymatically cleavable linker systems. We specialize in engineering linkers with tailored spacer architectures to modulate release kinetics and physicochemical properties, as well as enabling controlled release of challenging payloads.

Our capabilities further include late-stage functionalization of known payloads, de novo design and synthesis of novel payloads, and advanced strategies such as dual-payload ADCs and hydrophobicity optimization to support improved developability and pharmacological performance.

Enzymatic Assays for Payload Release Profiling

The design and optimization of linker-payloads, through lead selection, are driven by in-house enzymatic assays and complemented by in vitro and in vivo data provided by external clients.


SpiroChem has developed enzymatic assay to rapidly assess payload release kinetics from the payload-linker construct. This test enables fast screening of payload-linker constructs and accelerates decision-making over DMTA cycles.

From mAb to ADC Proof of Concept

Leveraging a portfolio of validated linker-payload systems ready for bioconjugation, SpiroChem enables rapid generation of ADC candidates without the need for extensive upfront payload or linker development. Combined with our bioconjugation expertise, we can produce and characterize a panel of ADCs to help clients evaluate different payloads, linker technologies, and conjugation strategies.
